產品描述：
MI培養基(MI Medium)是大腸菌和大腸桿菌顯色培養基，用于濾膜法檢測水中的大腸菌群。MI培養基是EPA 開發的，目前被批準用于檢測飲用水和其它形式的水中的大腸菌的檢測。優勢：1.快速。培養24h即可完成增菌和鑒定，無需驗證試驗。2.結果易讀。MI培養基是顯熒光培養基，在紫外光下，大腸桿菌菌落顯深藍色 ，易于與其它菌落區別開來。3.結果更精確。MI培養基可以直接用于大腸菌群計數，比基于統計學方法的MPN法和MTF法更精確。靈敏度為1 CFU/100 mL。
工作原理
MI培養基具有選擇性，抑制劑十二烷基硫酸鈉和脫氧膽酸鈉抑制非大腸菌的生長。生熒光化合物4-甲基傘 型酮-beta-D-半乳糖苷（MUG）和生色化合物吲哚-beta-D-葡萄糖醛酸苷環己基銨鹽（IBDG）分別用于檢測大腸菌群的beta-半乳糖苷酶活性 和大腸桿菌beta-β-葡萄糖醛酸酶活性。大腸埃希氏菌表達β-葡萄糖醛酸酶催化生熒光底物4-甲基傘型酮-β-D-葡糖苷酸(MUG)水解，釋放 熒光化合物。366nm波長的紫外光照射下菌落發出藍色熒光。大腸菌表達beta-半乳糖苷酶，水解生色底物IBDG，釋放藍色產物，使菌落顯藍 色。用途：

配制方法：
1. 稱取21.52g本產品，用1000ml去離子水浸泡10min。溫和加熱攪拌至完全溶解
2. 分裝。
3. 121°C滅菌15min。
4. 配制好的培養基必須避光保存。
實驗方法
35 ±0.5°C培養24h，觀察培養結果。1. Mix the broth by inverting the ampules two or three times. Pour the contents evenly over the absorbent pad and place the lid on the petri dish.
2. Filter the sample or diluted sample using a 47 mm white gridded 0.45 μm membrane filter, grid side up. Transfer the filter to the previously prepared petri dish using sterile forceps.
3. Place the filter, grid side up, onto the absorbent pad with a slight rolling motion. Check for trapped air under the filter and make sure the filter touches the entire pad. Replace the petri dish lid.
4. Invert the petri dish and incubate at 35 ±0.5°C for 24 hours.
5. Count the colonies. Use a 10 to 15X stereoscopic microscope, if necessary. E. coli colonies produce a deep blue color under ambient lighting. Coliform colonies fluoresce under UV light. No confirmation step is required.
